Yes, you need a carnet for Australia.
You'll need CTP (Compulsory Third Party insurance) - we obtained ours in Queensland - initially they said that they didn't issue CTP for foreign vehicles however relented once we talked to a supervisor. AA of Australia helped us navigate the uncertainties with this. If you ship into NSW you may have a problem because in NSW CTP is in the price of petrol, however when you leave NSW you'll need a certificate. At first we (mis) understood that we would have to get CTP for every state and ACT, however we found that the Queensland CTP covered all of Australia.
There are lots of stories on various fora that you need safety inspection and some other state stuff, but with the carnet, as long as the vehicle and bike are legal at home you don't need state inspections. The stories have to do with people importing vehicles (and registering them in Australia).
If your Defender is LHD you don't need a LHD sticker (it's not over the limit for this in terms of GVM).
You will not have to pay road tolls - foreign-registered vehicles are ignored.
