You can buy petrol from locals on the way at the road side, so do not worry... also, fill up from the people at the guest house on the Iranian side. You want to set off early, say 6am to ensure you get to Quetta before or around dusk.
It is best to do the crossing from Pakistan late afternoon, spend the evening at the guest house on the Iranian side (good food, hospitable but haggle over rates as always, no running water or electricity when i was there) and make that early start with a full tank.
The ride is quite pleasant really, with the main problem being the military check points... who usually want to spend to long talking to you! or offer to make you tea! just keep going as consistently fast as you can without over pushing it, as other traffic is worse for road manners than Iran... enjoy, but keep alert and whits about you on the long ride.
